Bacterial total RNA for RNA-seq library prep were quantified using Qubit RNA HS assay kit. This assay protocol is more specific than spectrophotometer based quantification. Quantification results are reliable and recommended for further downstream library prep including rRNA removal. Method can detect even picogram levels of RNA in the samples.

Bacterial RNA was diluted to 1:10. This 1:10 diluted samples was used at 1:200 dilution to measure RNA quantity using assay kit with manufacturer's instructions.

Tips

Run standards first and dilute origin RNA samples before making actual measurements

Results Summary

1:10 diluted samples with 1:200 dilution (working buffer of Qubit RNA HS assay kit) gave me 450 ng/microliter of Total RNA.
