Quantitative Western blots allow our lab to gain quantitative information regarding the amount of a protein in a given cell line. We use normalization Western blots to ensure we load the same amount of protein in each well. We chose the antibody from LiCor because we use their fluorescent imaging system.

Mouse Beta Actin, 1:1000, 8 hr incubation at 4 degree Celcius

Odyssey PBS Blocking Solution (Licor)

Secondary Incubation

Donkey anti-Rabbit 800CW, 1:10,000, 1 hr at room temperature

Tertiary Incubation

No tertiary incubation. Washed blot 3-times with PBS-T after secondary incubation before scanning

Detection

Flourescence detection with Li-Cor Odyssey system

Results Summary

The band for B-actin is visible at 45 kDa, directly between the 50 kDa and 38 kDa molecular weight standards.

Additional Notes

Try using the recommend amount of antibody before using higher concentrations, as higher concentrations may cause imaging difficulties
