In order to find out what cells play a roll in the proteoglycan (PG)-induced arthritis (PGIA) model, we identify the cell populations in the different tissue of PGIA mice by flow cyotemetry. In this experiment, we used anti-mouse CD62L as a naive T cell marker.
Flow Cytometry
Splenocytes and peritoneal cells
1X10^6 splenocytes or peritoneal cells were incubated with 0.5ul PE anti-mouse CD62L and other Abs in 50ul FACS buffer for 30 min at 4C.
Cells were blocked with 0.5ul CD16/CD32 for 10 min at 4C
