The aim of the study was to identify major leukocytes subsets in human peripheral blood. Additionally we were interested to identify leukocyte effector function. Since we used children in this study the goal was to use minimal amounts of blood. Therefore we performed multicolor flow cytometry and were in need of reliable antibody staining. Used antibody were first titrated to determine the exact dilution yielding a discriminatory staining and to reduce spectral overlap with other fluorochromes.

Primary Incubation

50ul of blood was incubated for 30 minutes at room temperature in the dark with the antibody. After this peripheral blood was lysed using the BD lysing buffer as indicated in the manual. Cells were washed three times with PBS and then resuspended in 300ul of FACS buffer and taken to flow cytometry.
