The aim of the study was to identify major leukocytes subsets in human peripheral blood. Additionally we were interested in identifying leukocyte effector function. Since we used children in this study the goal was to use minimal amounts of blood. Therefore we performed multicolor flow cytometry and were in need of reliable antibody staining. Used antibody were first titrated to determine the exact dilution yielding a discriminatory staining and to reduce spectral overlap with other fluorochromes.

Primary Incubation

50ul of human peripheral blood was stained with the antibody fro 30 minutes at room temperature. Then, cells were lysed with 1ml BD lysing solution. After three washing steps with 1 ml PBS, cells were resuspended in 300ul FACS buffer and taken to measure.
