Product Name:

Human/Mouse GLI‑3 Antibody

Catalog Number:

AF3690

The goal of this antibody was to test if Gli3 full length and repressor forms are expressed in NIH3T3 cells and if their levels change in response to Sonic Hedgehog ligand addition.

Experimental Design and Results Summary

Applications

Western Blot

Sample

NIH3T3 cells

Primary Incubation

1:200 dilution in blocking buffer for 16 hr at 4 deg

Blocking Agent

5% Milk powder in 1xTBS-T (0.1% Tween)

Secondary Incubation

1:5000 dilution (Donkey anti-goat IgG HRP) in blocking buffer for 1 hr at room temp

Tertiary Incubation

NA

Detection

ECL pico for 5 min

Results Summary

The antibody detects both both Gli3 full length (~200 kDa) and Gli3 repressor (~80 kDa) forms. After Sonic Hedgehog ligand addition for 24 hours, both Gli3 full length and repressor forms are degraded (as published in literature).

Summary

The Good

Detects both Gli3 full length and repressor forms

The Bad
