To estimate the Total Tumor Necrosis Factor present in bovine plasma, Bovine Total Tumor Necrosis Factor ELISA Kit was used which is a sandwich ELISA based method. The presence of TNF was detected by the appearance of colored product.
Tumor Necrosis Factors which are involved in apoptosis can be detected.
Bovine plasma
Blood from bovine is collected in EDTA coated tubes. After centrifugation, upper layer (plasma) was transferred into a fresh Eppendorf and bovine Total Tumor Necrosis Factor was detected in plasma according to the manufacturer’s instructions. 20 µl bovine plasma was dispensed in antibody coated 96 well plate, which was reacted with enzyme-linked antibody. After incubation, substrate was added and colored product was formed which was detected in ELISA reader.

The colored product after adding the substrate was placed in ELISA plate reader and absorbance was measured which was interpolated in the standard graph and the concentration of TNF was estimation
The kit protocol consumes less time and the results were repeatable.
