Liberase TL (thermolysin low) is a research grade enzyme that is used for dissociation of a broad range of tissues particularly mice. it contains both purified collagenase I and collagenase II in a precise ratio that works best for mouse tissues. I used this enzyme to dissociate mouse tumors followed by density gradient centifugation for harvesting tumor infiltrating lymphocytes. I got a good recovery of lymphocytes with just a 30 minute dissociation with liberase.

Protocol Overview

Incubate the tissue with enzyme for 30 minutes followed by manual agitation and desnsity gradient centrifugation. Collect the middle layer to isolate purified lymphocytes.

Prepare enzyme fresh everytime before use because of its short half life

Results Summary

The recovery of lymphocytes from tumor tissue was pretty good; enough cells to perform analysis by flow cytometry
