To estimate the cellular metabolic activity in Caco-2 cells, MTT Cell Proliferation Assay Kit was used.

Caco-2 cell line
Caco-2 cell lines when reached at its confluency, their viability and activity was measured by MTT assay kit. The kit contains MTT powder (125 mg) and cell based buffer (25 ml). Before the assay, MTT powder was completely dissolved by mixing properly in buffer by inverting the tubes. This resulted in yellow colored solution. This yellow color is light sensitive, therefore the solution was kept away from light. This assay is a colorimetric assay and in the presence of oxidoreductase enzymes the yellow color was reduced to purple color. Therefore, the greater the number viable cells, the greater the formation of purple colored product. MTT assays were done in the dark and performed according to the manufacturer’s instruction.

The purple colored product was measured on the plate reader and the results showed good repeatability.
