Company:

EMD Millipore

Product Name:

Anti-Sox9 Antibody

Catalog Number:

AB5535

The SRY/HMG box (Sox) family of transcription factors are expressed in early pancreatic development. Among these, Sox9 is expressed at its highest level in the early pancreatic epithelium. It has been shown that all pancreatic cells derive from Sox9 expressing precursor cells. Adult pancreatic ductal epithelium expresses Sox9.

Experimental Design and Results Summary

Applications

Immunofluorescence

Sample

e15.5 mouse pancreas region; paraffin embedded

Primary Incubation

1:500; 4 deg C; Overnight

Blocking Agent

5% Normal Donkey Serum in 1%BSA/PBS (w/v)

Secondary Incubation

1:1000. rabbit IgG-Cy2. 2 hours, Room temperature

Tertiary Incubation

NA

Detection

Fluorescence microscopy

Results Summary

Sox9 is easily detected in the pancreatic epithelium at e15.5. The counter-stain of Pdx1, a pancreas-specific transcription factor, was used to confirm the presence of pancreatic epithelium region.

Additional Notes

Antigen retrieval (TEG, pH9.0) was necessary to detect Sox9 and Pdx1 in paraffin embedded sections.
