This Biophotometer from Eppendrof was used to measure protein concentration in whole cell lysate. For this, relative absorbance of protein samples with respect to standards were measured using Eppendorf's biophotometer.

Protocol Overview

Cell lysate was prepared in RIPA buffer and 5microL was mixed with 1X protein estimation BCA Reagent. Solution was heated in water bath at 60 0C for 1hr to develop blue color. Similarly, standard BSA was prepared from a range of 2 to 18 microgram with a difference of 2. Absorbance of samples was measured at 562 nm wavelength in biophotometer and compared to that of standards.

Results Summary

Biophotometer had showed fast and accurate measurement of absorbance. No effect of an electromagnetic field was observed in sample readings. If the same solution was read twice, almost the same absorbance readings were observed.
