Whole cell protein lysate was resolved in 7.5% SDS-PAGE at 140 V using Bio-Rad PowerPac HC. Resolved proteins in SDS-PAGE gel were assembled in cassette of submerged western transfer unit, and proteins were transferred to the nicrocellulose membrane at constant voltage (94V) for 2 hrs. Further, transferred membrane was probed with specific monoclonal primary antibody followed by secondary antibody to detect p300 protein.

SDS-PAGE gel, PAGE buffer, nitrocellulose membrane, submerged western transfer assembly, Transfer buffer, primary and secondary Ab, etc.
Whole cell proteins were extracted in RIPA buffer, quantified and resolved in 7.5% SDS-PAGE. A constant potential difference of 80V was applied throughout stacking gel and 140V for separation. After SDS-PAGE, proteins were western transferred in nitrocellulose membrane using submerged assembly with a constant voltage of 94V throughout the run. Further, membrane was blocked in 5% skimmed milk and probed with p300 antibody.

Bio-Rad PowerPac HC had showed excellent separation of proteins in SDS-PAGE, indicated by absence of any streaking. Upon western transfer using same power pack, a high molecular weight protein p300 got efficiently transferred to nitrocellulose membrane. There were no abnormalities found related to western transfer which indicated precise smooth output voltage throughout the run.
