Whole cell extract was prepared from mouse cell lines. For this, growth factor treated or untreated cells were lysed in buffer and crude extract was centrifuged in a Sorvall micro centrifuge machine to pellet insoluble cellular debris. Supernatant collected was resolved in 10% SDS-PAGE.

Whole cell extract prepared in RIPA buffer, 1.5 ml centrifuge tube, micropipette, tips etc.
Cell culture was grown, serum starved and treated with specific growth factor. RIPA buffer was used to lyse cells, incubated in rocking platform for 15 min and centrifuged in Sorvall™ Legend™ Micro 21RMicrocentrifuge machine at 12000xg for 10 min maintaining temperature 4 0C. After centrifugation, supernatant was collected and proteins were estimated and resolved in 10% SDS-PAGE.
Always use rotor cover to avoid spillage and cross contaminations of samples.
Upon centrifugation of crude cell lysate at 12000xg for 10 minutes, gives a concentrated palate of insoluble cellular debris. The supernatants were clear and easy to transfer in a fresh tube.
