Mouse orthotopic syngeneic mammary tumors established by MVT1 cells were analyzed for the presence of T cells using APC anti-mouse CD3 antibody and flow cytometry. The tumors were pulverized to obtain a single cell suspension using Collagenase IV. Incubate the cells with 5% serum in TBS buffer for 30 min on ice. Remove the blocking buffer and incubate the cells with CD4 antibody diluted 1:100 in blocking buffer on ice for 30 min. Wash the cells 3 times with TBST. Fix the cells in 2% peraformaldehyde. Analyze percentage of cells present in tumors using FACS. The APC anti-mouse CD3 Antibody successfully detects CD3 positive T cells.
