We performed protein expression analyses in human podocytes regarding the generation of different factors involved in apoptosis regulation, signaling, cell structure maintenance and cell viability. Specifically phospho-specific antibodies were used to analyse the signaling activity normalized againt total protein expression (see Akt, AMPK and mTOR). In this experiment a a monoclonal Anti-Desmin antibody from Sigma Aldrich # D1033 was used (1:500) for Western blot.

Dilution 1:500, 16 hours, 4°C
5% Milk powder in TBST buffer
Dilution: 1:5000, 1 hour, room temperature
ECL - 5 min, room temperature
ECL detection
Different bands were detected after 5 min includng a desmin-band at 55 kDa (see figure).
Desmin was detected at 55 kDa
Several other bands were detected, too (Desmin Ab, 1:500)
This antibody is ok for Western blot analyses of desmin expression in human podocytes
