This antibody was used in a study designed to analyze the recruitment of stromal cells to the tumor in breast cancer mouse models. The goal was to study the presence of CD11b positive macrophages and myeloid derived suppressor cells in the tumor microenvironment. The APC conjugated CD11b antibody is easy to use and produces great results in less time.
Flow Cytometry
Primary tumors from spontaneous, orthotopic and xenograft breast cancer mouse models
The antibody was diluted to 1:200 and incubated for 30 minutes on ice.
2.5% fetal bovine serum and 2.5% BSA in TBS buffer
None
APC conjugated primary antibody was detected by FACS analysis
The CD11b successfully detect myeloid cells. Around 10-12% of total cells were CD11b+
Image caption: The tumor cells were pulverized and single cells suspensions were prepared. The cells were blocked in 2.5% FBS+2.5% BSA in TBS buffer. The cells were incubated with CD11b antibody (1:200 in blocking buffer) for 30 minutes on ice. The cells were washed with TBST for 10 minutes 3 times. The cells were fixed by 2% paraformaldehyde in PBS and analyzed by FACS
