The general aim of this project was to assess total protein translation attentuation in a variety of treatment groups using total and phosphorylated eIF2alpha, a downstream component of PERK. To do this we compared phosphorylated eIF2alpha to total using western blotting.

Primary Incubation

1:500 antibody, 0.1% tween, in 1:3 li-cor odyssey block: TBS overnight at 4 degrees C.

Blocking Agent

Li-Cor Odyssey Block for 1 hour.

Secondary Incubation

1:15,000 Li-Cor IRDye 800CW, 0.1% tween, 0.02% SDS, in 1:3 odyssey block: TBS at room temperature for 1 hour

Results Summary

Phosphorylated eIF2alpha indicates translation attentuation. This antibody detects endogenous levels of eIF2alpha protein when phosphorylated at S51.
