I wanted to use BMP4 as a positive control for my stem cells since I was trying to identify new compounds that can induce differentiation in my human stem cells.

Cells were seeded in laminin coated wells of a 96 well plate. After allowing them to adhere for one day, they were treated with 100ng/ml of BMP4. 7 days post-treatment they were stained for GFAP and the number of GFAP positive cells were counted in the vehicle control DMSO and the BMP4 treated cells. Counts were normalized to untreated cells
Remember to check solubility of the compound.
Cells that were treated with the BMP4 gave a clean result as a larger proportion of the stem cells showed positivity for the differentiation marker of choice
