Following vaccination, exhaustion of the induced T cells is an important consideration. Exhaustion of the T cells can result in poor T cell functionality and poor vaccine efficacy. PD-1 is a marker of T cell exhaustion, and thus staining for the marker on T cells in PBMCs is of importance. This antibody successfully stains for PD-1.

Experimental Design and Results Summary

Applications

Flow Cytometry

Sample

Murine PBMCs from blood draw.

Primary Incubation

0.5 ul Antibody per 2x 10^6 cells, diluted in MACS buffer in final volume of 50ul.

Blocking Agent

MACS buffer. Incubated for 30 mins at 4C in the dark.

Secondary Incubation

Results Summary

This PD-1 antibody successfully stains PD-1+ cells from murine PBMCs. It allows the distinction of PD-1+ and PD-1- cell subsets from a heterogenous population, as well as the MFI, or the mean amount of PD-1 expression per cell.
