To stain the neutrophil population in mouse peritoneal cells, mice were pretreated with a drug or PBS control, followed by injection with LPS via i.p. Peritoneal cells were then collected 4 hr later. Cells were blocked with Fc blocking antibody and then stained with FITC-anti mouse Ly-6G and analyzed by FACS. The results showed that a specific population of cells was increased in control but not treated mice.

We used Fc blocking antibody before staining the target marker in FACS and the results showed that antibody staining was very specific, suggesting that Fc blocking antibody worked very well. FACS results showed that drug treatment inhibited LPS induced neutrophil recruitment in the peritoneal cavity.
