To analyze neutrophil recruitment, mice were pretreated with PBS or a drug (LF fraction 10). 48 hours later, mice were injected with LPS via i.p. and peritoneal cells were collected 4 hours later. Cells were blocked with Fc blocking antibody and then stained with FITC-anti mouse Ly-6G. Cells were analyzed by FACS. We found that treatment inhibited LPS induced neutrophil recruitment in the peritoneal cavity.

Experimental Design and Results Summary

Applications

Flow Cytometry

Sample

Mouse freshly isolated peritoneal cells

Primary Incubation

Anti-mouse CD16/32 Antibody

Blocking Agent

5% FBS in PBS

Secondary Incubation

FITC-aM Ly-6G

Tertiary Incubation

NA

Detection

Flow cytometry Canto II

Results Summary

The drug treatment inhibited LPS induced neutrophil recruitment in the peritoneal cavity. While PBS treated mice showed increased neutrophil activity. The antibody is specific and sensitive for staining the neutrophil marker.

Additional Notes

Antibody incubation on ice for 15-30 min is enough

Summary

The Good

The antibody is specific and sensitive in one step staining.

The Bad

None

The Bottom Line

This antibody is good for FACS experiments for staining a neutrophil population. I have not yet combined it with other antibodies to costain cells.
