The aim of the experiment was to test the mouse genotype for Par1 allele. For this, mouse tail DNA was isolated using Direct PCR (Tail), PCR was performed using specific primers and visualized in ethidium bromide containing agarose gel.

Mouse pups of 3 weeks age were anesthetized with isofurane and 1mm of mouse tail at tip was excised using sterile scissors. Tail was directly placed in 200 microlitres of Direct PCR (Tail) reagent containing 6 microgram of Proteinase K. Tubes were incubated overnight at 55C followed by 85C for 2 hours. Tubes were centrifuged at 2000rpm for 1 min and one micolitre of supernatant was run for PCR with specific primers.
