I screened a lung cDNA library for a potential ESAT6 binder using bacterial two hybrid system. We used X-gal selection plate to screen for potential interacting partners.

I cloned ESAT6 protein in pBT vector and cloned a lung cDNA library in pTRG vector then cotransformed both vector mixes in a reporter strain and plated on X-gal indicator plate at 30 degrees for 36 hours.

Found positive interacting peptides, namely HCL2 and SL3, with mycobacterial ESAT6 protein.
Serves as a good indicator for bacteriological assays.
