CD25 or IL-2 receptor alpha is only expressed on Treg cells and transiently expressed on activated T cells. I have never performed CD25 staining on the brilliant violet channel; so wanted to test this antibody as other channels were occupied. The stain using BV421 was as good as other bright fluorophores like PE, FITC etc. as observed in my previous experiments.

Results Summary

There is a clear CD25 positive population on ex vivo isolated lymphocytes from a mouse spleen. The frequency of positives is physiological. The populations are not as distinct as observed with other bright fluorophores.
