Protocol Overview

Take 100micro liter of staining buffer and then add each flurochrome-conjugated antibodies and label them separately. Add 1 drop of the BD comp beads and Anti-Rat Tg,K beads to each tube and incubate at dark for 30 min and centrifuge at 200xg for 10 min. Wash with FACS buffer once and suspend again with FACS buffer and use as a compensation control.

Tips

Before using the fluorochrome labelled antibodies, it is better to a titrate the reagent and determine the concentration.

These compensation beads are essential for doing all flow cytometry studies to reduce background fluorescence from the positive and negative stained populations.
