Purified Mouse Anti-Human HIF-1α

I wanted to test the efficacy of HIF 1 alpha inhibitors in cells. I artificially induced upregulation of HIF 1 alpha using Cobalt chloride and then added my inhibitor of choice. 6 hours post-incubation I made lysates of the cells in RIPA buffer and loaded 10ug of protein on a precast Biorad Protean mini gel. I then performed a western blot for HIF 1 alpha using the antibody.

Results Summary

The antibody worked exceedingly well and did not require optimization. It did not detect any HIF in the control GSCs, detected a large amount of HIF1 alpha in the GSCs treated with cobalt chloride and did not detect any HIF1 alpha in the inhibitor treated cells showing the high specificity of the antibody for the protein
