This antibody was used in our study of proteasome inhibitor resistence in multiple myeloma published in Cancer Cell (Leung-Hagesteijn et al, "Xbp1s-negative tumor B cells and pre-plasmablasts mediate therapeutic proteasome inhibitor resistance in multiple myeloma", 2013 Sep 9; 24(3):289-304). The antibody gave excellent results in ICC/IF staining in primary patient bone marrow samples and multiple myeloma cell lines.
Immunofluorescence
cd138+ primary patient bone marrow cells, multiple myeloma cell lines JJN3 + XBP1s-GFP and RPMI-8226 + XBP1s-GFP
1/100 overnight at 4oC in 10% goat or donkey serum/PBS
10% normal goat or donkey serum in PBS
1/1000 Alexa Fluor 647 donkey or goat F(ab")2 anti-mouse IgG (H+L)

Zeiss AxioImager Cy5 cube 700 ms exposure
Specific detection of XBP1s in GFP-positive XBP1s/GFP transfected cells. Minimal background in untransfected cells. Strong staining in cd138+ primary patient bone marrow cells.
Specific staining of spliced XBP1 (XBP1s)
Excellent reagent for ICC/IF of XBP1s splice variant (difficult to find).
