I am comparing the size of the endoplasmic reticulum to the size of the whole cell in a variety of treatment conditions. The ER was successfully labeled using a GFP transfection probe but I needed to have a marker for the outside of the cell to accurately compare the size of the organelle to the whole cell.

Protocol Overview

After cells were confluent, the slide was place in the staining solution (2.5ug/mL or 1.25ug/mL) for 5-10 minutes at 37 degrees C and rinsed three times with PBS. Cells were then fixed with formaldehyde and mounted using vectashield mounting medium.

Tips

We used their suggested concentration to begin with and it was way too much.

Results Summary

The orange color bled into the green channel. I later found out this was because the microscope band pass was wide enough to allow for bleeding into the yellow-green emission. After looking over the product sheet again, it is NOT suitable when combining other organelle stains because it is hydrophobic and has permeability issues. It did work to stain the entire cell, however, so anyone having to measure that alone should have great success.
