Equal amounts (10 micrograms) of protein was resolved in SDS-PAGE and western transferred onto PVDF membrane. Transferred blot was probed for low abundance proteins, treated with ECL, and X-ray films were developed. ECL reagent from Thermo Scientific was selected because of its cheap cost and easy availability

Mammalian cells were grown after which proteins were isolated, resolved in 10% SDS PAGE and immunoblotted for low abundance proteins. Primary antibody and HRP-conjugated secondary antibody were respectively taken in 1:1000 and 1:5000 dilution. Probed blot was incubated in ECL solution for 1 min at RT. X-ray film was exposed with blot for 1-5 seconds and developed.
