I used this BglII restriction enzyme along with EcoRV for the cloning of my gene of interest into a plasmid vector. Subsequently, I also used this enzyme for colony screening. This enzyme completely digested my PCR products and vector which helped in getting my clones in the first attempt.

I used this BglII restriction enzyme along with EcoRV for the cloning of my gene of interest into a plasmid vector. I used 5U of enzyme for digestion of 1 microgram of DNA at 37c for 2h.
Whenever we do a double digestion using BglII and some other enzyme, it's better to select an appropriate buffer which is compatible for both enzymes instead of using NEB buffer 3.1 supplied with BglII. We can also extend digestion up to overnight.
