We wanted to examine the localization of Foxp3+ T-cells in healthy vs diseased mice, so we turned to this antibody for immunofluorescence identification of Tregs.

Experimental Design and Results Summary

Applications

Immunofluorescence

Sample

Pre-fixed frozen mouse small intestine tissue section

Primary Incubation

1:100 in 1% BSA in PBS overnight at 4 degrees

Blocking Agent

5% goat serum in 1% BSA in PBS

Secondary Incubation

Goat anti-rat AlexaFluor647 was used at 1:200 in PBS and was incubated on the section at 1 hour at room temperature in the dark

Tertiary Incubation

NA

Detection

Zeiss LSM 700 confocal

Results Summary

Foxp3+ T-cells were clearly identified/stained in the mouse tissue section. The stain clearly stains the nucleus of the T-cells.

Additional Notes

CD3 staining is shown in red while the Foxp3 staining is shown in white

Summary

The Good

Stains Foxp3+ T-cells on tissue sections well, and it exhibits the expected nuclear localization

The Bad

Nothing really to mention (though there did seem to be a bit of background/nonspecific non-nuclear staining)

The Bottom Line

Great antibody for identifying Foxp3+ T-cells via immunofluorescence
