The aim of the experiment was to test the role of growth factor on F-actin stress fiber formation in mammalian cells. Phalloidin, Rhodamine 110 conjugate from Biotium was used to stain F-actin stress fibers.

Experimental Design and Results Summary

Application

Detection of growth factor induced F-actin stress fiber formation.

Starting Material

DPBS, 4% paraformaldehyde, 0.5% triton X-100, 0.3% BSA, DAPI, Gold antifad mounting medium.

Protocol Overview

Mammalian cells were grown on cover slips, starved for 12h and treated with growth factor. Cells were fixed in paraformaldehyde for 15 min at room temperature, permeabilized with triton X-100 for 10 min and blocked 1h with 0.3% BSA. Cells were incubated with 6.6mM Phalloidin (1:2000) for 1h, washed with PBS and mounted with slide. Cells were observed in epi-fluroscent microscope with excitation wavelength of 502nm and images were captured. Red colour was artificially selected using microscope software.

Use Phalloidin diluted in PBS (1:2000)

Results Summary

Got very good staining of F-factor stress fibers.
