We had previously used Millipore antibodies to determine the levels of RAGE in mouse lungs but were not able to get clear bands for target protein. Here we are using Abcam RAGE antibody to determine the level of RAGE in different transgenic mouse strains. This product was also used to determine the levels of RAGE in human gingival epithelial cells, but with less clear results.

Experimental Design and Results Summary

Applications

Western Blot

Sample

Whole tissue lysates from the periodontal area

Primary Incubation

1:1000 in 5% milk

Blocking Agent

5% Milk for 1 hr

Secondary Incubation

1:10,000 dilution anti-rabbit conjugated with HRP secondary antibody

Results Summary

In mice, we observed a clear RAGE target band at ~ 42 kDa. Non-specific: faint bands above ~60, kDa and above. In human tissue we observed three different bands (faint band ~42, prominent band at ~ 30 and faint band ~17 kDa) which was different from that mentioned in abcam data sheet (~42 kDa).

Summary

The Good

Defined target band in mouse tissues

The Bad

Some nonspecific bands are observed above the target bands in mouse model and not sure if the target band is RAGE in human model since the size differs from Abcam data sheet.

The Bottom Line

Works well in mouse tissues.
