For mucosal sampling of murine vaginal vaults, for either antibody or cytokine analyses following vaccination for example, an establish method is vaginal washing with PBS. This is performed with a positive displacement pipette, yet the tips used are of upmost importance. They need to be of the right size, dimensions and hold adequate volume as well as be accurate. For the Gilson Microman M100, the optimum tips for this use are the capillary piston tips (CP100ST).

Protocol Overview

In combination with the Gilson Microman M100 positive displacement pipette, these CP100ST capillary piston tips are used to take up 33 ul PBS, and flushed within the murine vaginal vault. There are 3 flushes of 33 ul, resulting in a final sample volume of 100 ul. These tips are discarded after each individual mouse to avoid cross contamination.

Tips

Ensure the pipette is pushed all the way through before attaching the capillary piston tips, and ensure the tips click onto the end of the pipette prior to aspirating media.

Results Summary

When used correctly in combination with the Gilson Microman M100 pipette, the mucosal sampling is reproducible, quick and minimally invasive, yielding mucosal samples from which ELISAs can be carried out. Murine total IgG and IgA concentrations are consistent between mice and groups when using these tips.
