This is very simple to use and it's a very reliable reagent for mouse genotyping
Mouse genotyping.
Mouse tissue from the tail or ear.
Genotyping protocol Day1: 1. Cut the tail for about 0.5-1.0cm, tail tag the mouse, and write the records. 2. Add 200µl Directly PCR lysis buffer and 10 µl proteinase K (20ng/ml), then incubate at 65 C overnight. Day 2: 3. Heat samples at 85 C for 45min to inactivate proteinase K. 4. Genotyping PCR µl 5X Gotaq PCR Buffer 4MgCL2(50 mM) 0.8dNTP(10mM) 0.5Primer mixture, Forward and reverse, 1µl /each oneGotaq Polymerase 0.2Tail DNA 2dd H2O 11.5Total 205. Run 2% or 3% Gel.
No need to mix the tissue after heating with reagents overnight

Works well.
We used this for the tail from the wean pups, but it also can be used for genotyping adult mouse ear or liver tissue, it's a very good reagent for mouse DNA extraction.
