Red blood cell lysis buffer

This buffer is intended to remove red blood cells from cell suspensions obtained from blood or secondary lymphoid organs. The incubation times are longer than comparative products and even after two rounds of lysis, I saw significant amount of Red cells in my suspension.

Protocol Overview

Pellet cells and resuspend in 2-3mL of red blood cell lysis buffer. Incubate at room temperature for 5 minutes followed by wash with appropriate buffer. The cells are ready for downstream applications

Tips

When working with blood, perform lysis twice with washes in between

Results Summary

Poor efficiency of lysis was observed and comparable products have better efficiency with shorter incubation times.
