Our lab is interested in the role of antigen presenting cells during oral tolerance induction. We analyze the phenotype and function of dendritic cell subsets after tolerance induction.

Results Summary

Very bright staining for CD11c. As activated T cells, B cells and NK cells also express CD11c we used CD3-PE/CD19-PE/NK1.1-PE as a "dumping channel" to avoid false negatives. CD11c staining shows 2 populations: CD11c high and CD11c low. CD11c high cells are conventional dendritic cells, CD11c low are mostly plasmacytoid dendritic cells.

Additional Notes

It is recommanded to digest organs using Collagenase D and DNase I before staining for dendritic cells. Dilution used is 1/300 in 50ul per sample.

Summary

The Good

Very bright staining. For a PE-Cy7 coupled antibody it is remarkably stable over time and even after fixation.

The Bad

Don't use too much. It is very bright.

The Bottom Line

Great antibody. With this antibody you can make good use of your PE-Cy7 channel.
