Our current project is all about identification of affected proteins and their levels upon specific treatment of mammalian (HeLa) cells. Hela cell lysate of 25 ug protein was loaded onto SDS gel (4%-12%) and MES was used for electrophoresis. Major bands appeared at ~90 kDa and ~50 kDa.
Western Blot
Treated and untreated Hela cell lysates, 25 ug protein
1:500 overnight at 4 degrees Celsius
Blocker=Skimmed milk, Buffer= 1XTris Buffer Saline+0.1 % Tween 20, 2hrs at RT
1:20,000 HRP-conjugated secondary, 60 minutes at room temp

The antibody pretty much fulfills the need, the only thing that bothered me is the appearance of a major band at ~100 kDa. Someone who has detailed knowledge of FASL might tell better why this band comes out.

The good thing is that at least you see your desired band
The bad part is you should not get confused with the intense 90 kDa band
It works and identified the protein in Western Blot
