This antibody was used to perform immunofluorescence microscopy on HeLa cells transfected with a V5 containing construct. Several anti-V5 antibodies from different host species were used. D3H8Q was used in parallel with several mouse anti-V5 antibodies. D3H8Q gave minimal background fluorescence and strong immunoreactivity for epifluorescence microscopy.

Experimental Design and Results Summary

Applications

Immunofluorescence

Sample

HeLa cells transfected with anti-V5 containing construct

Primary Incubation

D3H8Q (rabbit anti-V5) incubated 1 hour in PBS 1% + normal goat serum +0.1% Triton X-100 at a dilution of 1:500

Blocking Agent

PBS 1% + normal goat serum +0.1% Triton X-100

Detection

Epifluorescence microscopy, Nikon

Results Summary

This antibody worked well in this application. The staining was similar to several mouse anti-V5 antibodies used. The use of rabbit anti-V5 antibodies can expand multicolor applications with the use of different host secondary antibodies.

Additional Notes

This antibody also worked in SW480 cells with similar results. Different Alexa Fluor conjugated secondaries also produced similar results.
