We were assessing Aryl hydrocarbon receptor activation in bone marrow derived dendritic cells. Precursor cells were isolated with Lympholyte M from whole bone marrow suspensions and grown in the presence of GM-CSF for 5-7 days. We then used flow cytometry and RT-PCR to assess changes in dendritic cells after AhR activation.
Flow Cytometry
BD Comp Beads, and Fluorochrome conjugated antibody
Add 1 drop of BD CompBeads Negative Control to FACS tube, plus 1ul of antibody. Incubate for 5 minutes, and add buffer up to desired volume (200-300l).
Change forward and side scatter to get a clean, tight population.
BD CompBeads Negative Control gives a clean, small population to gate on for collection, and has never shown positive for antibody.

This is a great negative control bead for flow cytometry.
