Macrophages can be identified in tissue cross-sections via the CD68 intracellular marker. We used this product along with the use of Cell Signaling Tech's AlexaFluor488 (Catalog No. 4416S) secondary antibody to obtain remarkably excellent quantifiable images of various tissue cross-sections.

Primary Incubation

1/100 dilution at 4 deg C in the dark overnight

Blocking Agent

Blocking buffer used was PBS with 0.05% Tween 20, 1% BSA, 5% Host serum and 0.3 M Glycine

Secondary Incubation

Anti-rat Alexa Fluor 488 (Catalog No. 4416S from Cell Signaling Technologies), 1/200 for 1 hour at room temperature in the dark

Tertiary Incubation

Used Hoechst 33343 for nuclear staining

Detection

FITC filter in any standard epifluorescence microscope

Results Summary

As seen in the image, the overlay indicates positive CD68 staining after subtracting using the IgG control staining around cells (made visible via the nuclear stain - Hoechst 33342)
