Our aim is to maintain the breeding colony of our laboratory mice which is a KO mice. Specifically, our goal is to screen the KO, WT and heterozygous group of mice. We tried with different methods for the genotype but we found Mouse Direct PCR Kit (For Genotyping) is cost effective for fast preparation of genomic DNA and PCR amplification. On the same day we got the PCR results from 51 different tail sample. I would recommend this kit if you have to do the genotype on a small or larger scale.

The Buffer L and Protease Plus rapidly digest mouse tissue to release intact genomic DNA which can be used directly as the template for PCR amplification. The PCR product was directly loaded in the gel and visualized.
Key points: 1. L buffer should be stored at 40c, 2. Digestion should be 30-45 min., 3. I used 4µl of genomic DNA in my PCR reaction, 4. Annealing temp is important

We got the specific band for the WT, KO and heterozygous mice. The results are accurate and it was easy to separate the mice based on their genotypes.
