Retroviruses are critical in most therapeutic and gene transduction experiments. To optimize their performance, quantifying their titer post production and purification as well as in in vivo experiments can be time consuming. Using the SYBR green PCR master mix reduced the time used to whip up multiple PCR reaction tubes by >10-fold, especially in the initial phase of protocol development wherein different concentrations of primers and starting cDNA template were to be optimized.

Protocol Overview

Simple mixing of reagents is required as per manufacturer's instructions. The master mix is supplied at 2X concentration. Optimization of primers and cDNA amount is required and is independent of this product.

Tips

Clean lab bench without genetic contamination is necessary.

Results Summary

Efficient (~100%, slope=(-3.29 - -3.34) amplification is obtained upon using this PCR master mix at 2-fold dilution series (top, attached image), 5-fold dilution series (middle) and 10-fold dilution series (bottom). Blue dots show the standards.
