Visualization of gene therapy vectors is critical for quantitative evaluation of their functional performance in various cells. Here, we show that the lipid formulation of DOTAP, DOPE (with up to 3% of the fluorescent lipid 18:1 Liss Rhod PE) when combined with retroviral particles can be efficiently observed using fluorescence microscopy up to 4 h post transfection.

Protocol Overview

Simple mixing protocols were used wherein pre-determined number of retroviral particles and synthetic liposomes formulated with the fluorescent lipid were mixed and incubated for 2 h at 4 deg C. See paper for more details - http://www.ncbi.nlm.nih.gov/pubmed/25883029

Tips

Did not use 6 months post purchasing the fluorescent lipid. Use fresh and aliquot as needed.

Results Summary
