I am interested in postprandial change in TG levels after a co-ingestion of yogurt and a high calorie meal. Hourly blood samples were collected and centrifuged to obtain plasma samples. TG was measured by this colorimetric kit on a 96 well microplate.

Protocol Overview

1. Add calibrator (standard) and plasma sample (no need to dilute). 2. Add color reagent A, mix and incubate at 37 C. 3. Read at 600 nm. 4. Add color reagent B, incubate at 37 C. 5. Read again.

Tips

The kit used two points to make the standard curve, but I felt more conformatable making a standard curve based on more points. The volume of each reagent can be adjusted proportionally, depending on the maximum volume of the plate.

Results Summary

The final absorbance was calculated by subtracting the first reading from the second reading. Then make a linear standard curve. The unknown sample concentration was determined by the linear equation.

Additional Notes

The multi-lipid calibrator needs to be purchased additionally.
