Anti-S1PR1 biotinylated aptamers were used to stain white blood cells. Aptamers were first incubated with Streptavidin APC and then added to cells. Streptavidin APC didn't show any significant binding to white blood cells without aptamers.

Streptavidin APC was incubated with biotinylated aptamers in DPBS buffer for 25 min before aptamers were used to stain white blood cells

Tips

Heat up aptamers to 95C and cool down before incubation with streptavidin

Results Summary

Streptavidin APC by itself didn't bind to white blood cells. S1PR1-aptamer-biotin-streptavidin complex staind the same population as anti S1PR1 Abs
