Our laboratory selects aptamers for various cell receptors. To test the specificity of aptamer binding to white blood cells or transfected HEK-293 or MDA-MB-231 we use receptor specific antibodies. Aptamers can be labeled with fluorescent dyes or biotinylated. To detect biotinylated aptamers conjugated streptavidin is used. Here, white blood cells were stained with propidium iodide and S1P1 Abs to perform screening of anti-S1P1 aptamers.

Summary

The Good

Abs bind well

The Bad

Hard to analyze data unless additional Abs are used for other population-specific CDs

The Bottom Line

It is highly suggested to double stain white blood cells with an Ab of interest to identify which population of white blood cells is stained the most.
