The detection of mannose receptor in peritoneal macrophages (surface only) upon drug incubation was vital to our study. For this, we used a dual-antibody labeling strategy in conjunction with a pan macrophage marker for flow cytometric detection of alternatively activated macrophages. Primary Incubation

Dual-labeling strategy with co-incubation of antibodies (F4/80 + CD206). For F4/80 (1 ug/100,000 cells, 30 minutes, 4 deg C in dark)

Blocking Agent

Standard Fc block solution (Affymetric eBioscience - Cat No. - 16-0161-86)

Secondary Incubation

For CD206 (same as F4/80 as above)

Tertiary Incubation

None

Detection

MoFlo Astrios (BD), FITC channel

Results Summary

The detection of CD206 was rather easy relative to the isotype (shown in grey in the image). Primary gating with F4/80 was done to confirm that only macrophages with CD206 expression were analyzed.

Additional Notes

FITC signal may be weak. Use of better fluorochromes may enable better analysis

This antibody can be used for detection of alternatively activated macrophages. Typically surface expression may not be the best way to analyze these sub-type of macrophages. However, if only surface expression is of interest, this product works very well.
