I used this CoCl2 to mimic hypoxia in primary epithelial cells without having to put them in a low oxygen environment to try and understand the role hypoxia plays in different cell functions.

Protocol Overview

Make 100mM Cobalt chloride stock solution and then dilute 1:100 in cell culture media. Add media to cells and then incubate for 48 hrs and then measure HIF-1a protein levels from cell lysates

Tips

Be sure to dilute your stock solution because too much can be toxic

Results Summary

After adding cobalt chloride, HIF-1a protein was dramatically increased in Hela lysates compared to no cobalt chloride control.
