We routinely use the BD LSR II for multiple color staining, generally around 10 colors at the same time.

Normally, we stain 1 million cells in 50ul of staining buffer with titrated Fluorescence conjugated Abs together with cell viability dye for 20 minutes on ice and run the samples with the machine.
Be aware of the fluid tank, it won't automatically stop if the fluid is gone which will damage the membrane in the filter.
Can detect more than 10 colors at the same time, both facs tubes and 96-well plates can be used to run the samples. Both beads and cells can be used for compensation. Adjustment of compensation and data analysis is best achieved in FLOWjo.
Needs to warm up at least 30 minutes to run samples.

Way too big, takes a lot of bench space, file naming method is not handy, can't change name when accquiring data.
